The story takes place in the small town of Pascagoula, Georgia, where the local church choir is about to participate in a national competition called "Joyful Noise." However, just at this critical moment, the choir's conductor suddenly passes away, leading the choir to the brink of collapse without leadership. The church appoints a woman named Rosa (Queen Latifah) as the new conductor. Rosa is very traditional and wants everything to follow the usual norms, while the late conductor's widow, Sparro (Dolly Parton), believes she should take over the choir and try a new style.
