Guy (John Cassavetes) has a frail-looking wife, Rosemary (Mia Farrow). Since they moved into their new apartment, Guy has been getting along splendidly with an elderly couple next door. However, Rosemary is not particularly fond of them, and she feels increasingly uneasy about a series of strange events. Rosemary sees a woman commit suicide, and under the increasingly eerie atmosphere of her life, she starts having terrifying dreams. In her nightmares, a hairy monster assaults her, and shortly after waking from these nightmares, she learns that she is pregnant. The elderly neighbor comes to take care of Rosemary and her unborn child; however, Rosemary gradually begins to suspect that all the bizarre occurrences are tied to this seemingly benevolent neighbor and even her own husband! They are conspiring against Rosemary's baby. Are Rosemary's fears mere delusions, or is there something sinister happening? When she sees that her newborn's eyes have no pupils, Rosemary realizes that the devil's presence is close at hand.
